PCR Problems

I laugh at it so much because I understand what it’s like to work so hard and yet get a negative result, but actually performing PCR and getting a negative result… I still laugh but I’m crying on the inside.

I performed my first PCR experiment for my molecular biology lab course, and damn, I was only able to get the DNA ladder to show up:bio-335l-2018-03-02_18h15m39sneomi1.jpg

I expected these results since my transformation results were not expected (i.e. all my colonies were blue for my positive control instead of white, there was only one colony for the actually transformation, and got nothing in my negative control (no colonies whatsoever). So it hasn’t been the best couple of weeks for my experiments.

On the bright side, at least I know my transformation and PCR sucked. This one guy in my lab forgot to add the Safeview dye (a dye to let you see your bands), so his gel had nothing on it when he was getting the image.

*Sigh* Hopefully my transformations and PCR experiments end up successful in the future if I ever perform them.
